Effects of Aging and Calorie Restriction on the Extracellular Matrix Proteins in the Rat Tissues

Abstract
The effects of aging and calorie restriction (CR) on the extracellular matrix (ECM) proteins in the testis, kidney, liver and lung were investigated. Fischer 344 rats, ages 6, 12, 18, and 24 months, fed ad libitum (AL) and CR diets were utilized in this study. The interstitial collagen type I protein level was increased in the testis and kidney and lung with aging. But CR prevented these age-related increases in the testis and kidney. Fibronectin, the member of both interstitial matrix and basement membrane, was increased in the testis, kidney and liver with aging. CR did also prevent the age-associated increase. In the testis, the protein level of collagen type IV was decreased in 18 and 24 months of age. But in CR rats, the type IV collagen protein level was maintained until 24 months of age. In the kidney, aging and CR didnt have any significant effect on the protein level of collagen type IV. In the liver, the protein level of collagen type IV was increased with aging, and CR prevented this increase. In the lung, the collagen type IV was not detected in our experiment. These results suggest that the accumulation of collagens and non-collageneous composition of ECM is a common process with aging. But CR modulated these age-related changes in the composition of ECM proteins during aging process.
